中文摘要:目的:建立雌性大鼠去卵巢骨质疏松模型,模拟妇女绝经后骨质疏松,探讨低频脉冲电磁场对一氧化氮含量、一氧化氮合酶活性变化的影响,及其与骨重建的关系。方法:实验于2003-08/11在上海体育学院解剖教研室完成。选取3月龄健康纯种雌性SD大鼠24只,随机分为假手术对照组、去势对照组、低频脉冲电磁场组,8只/组。①去势对照组、低频脉冲电磁场组大鼠完整摘除两侧卵巢,假手术对照组找到卵巢后割取与卵巢等量脂肪。②低频脉冲电磁场组大鼠给予低频脉冲电磁场干预,电磁场频率为50Hz。将两只大鼠尾部相对置于螺线管中部,头部面向管的两端,治疗1h/d,5d/周,共12周。③各组大鼠处死前不同时间点进行2次荧光标记,以动态观察两次注射期间骨形成的情况。④第12周末治疗结束后取材,测定各组血清与骨匀浆中一氧化氮含量及一氧化氮合酶活性的变化,对腰椎骨进行骨形态计量学指标测定。结果:实验选取SD大鼠24只,全部进入结果分析。①磁场干预结束后各组大鼠体质量、子宫重量和血清雌激素含量测定结果:与假手术对照组比较,去势对照组、低频脉冲电磁场组大鼠体质量均明显升高(P<0.05),子宫质量、血清中雌激素含量均明显降低(P<0.05),而此两组之间差异均无显著性意义(P>0.05)。②磁场干预结束后各组骨形态计量学指标的测定结果:与假手术对照组比较,去势对照组、低频脉冲电磁场组骨小梁相对体积比、骨小梁平均骨壁厚度、皮质骨壁平均厚度均明显降低(P<0.05);骨小梁形成表面、骨小梁吸收表面、骨矿化沉积率、四环素标记荧光周长百分率、破骨细胞数量均明显升高(P<0.05)。③磁场干预结束后各组血清和骨匀浆上清液中一氧化氮含量及一氧化氮合酶活性的测定结果:低频脉冲电磁场组一氧化氮含量及一氧化氮合酶活性均较去势对照组显著升高(P<0.05)。结论:低频脉冲电磁场治疗可使去卵巢大鼠一氧化氮含量及一氧化氮合酶活性增高,从而抑制破骨细胞的骨吸收,降低骨重建的高转换率。

外文摘要:AIM: To establish ovariectomized rat models with osteoporosis and imitate postmenopausal osteoporosis in females, explore the effect of low frequency pulse electric-magnetic field on content of nitrogen oxide and activity of nitricoxide synthase (NOS), and the relationship with bony remodeling. METHODS: The experiment was performed at the Department of Anatomy, Shanghai University of Sport between August and November 2003, Totally 24 3-month healthy pure breed female SD rats were selected and assigned randomly into sham operation control group, gonadectomy control group and low frequency pulse electric-magnetic field group with 8 rats in each group, ①Bilateral ovaries were extracted from rats in the gonadectomy control group and the low frequency pulse electric-magnetic field group, The same volume of fat was isolated from the rats in the sham operation control group after finding out the ovaries, ②The rats in the low frequency pulse electric-magnetic field group intervened with low frequency pulse electric-magnetic field, The frequency of electric-magnetic field was 50 Hz. Tails of two rats were near to each other laid in the center of solenoid, Their heads faced to the edge of the solenoid, treating for 1 hour daily, 5 days in a week, totally for 12 weeks, ③Two fluorescent labelings were performed at different time point before being killed in rats of each group. Bone formation was observed dynamically in the two injections. ④Materials were drawn at the end of the 124 week to determine the changes of the content of nitrogen oxide and activity of NOS in serum and bone homogenate, and also detect the index of bone morphometry of lumbar vertebra. RESULTS: Totally 24 experimental rats were involved in the result analysis. ①Assaying findings of body mass, womb weight and content of serum estrogen of rats in each group after magnetic field intervention: Compared with the sham operation control group, body mass of rats in the gonadectomy control group and low frequency pulse electric-magnetic field group significantly increased (P 〈 0.05). Womb weight and estrogen content in serum obviously decreased (P 〈 0.05). There was insignificant difference between the two groups (P 〉 0.05). ②Assaying findings of index of bone morphometry of each group after magnetic field intervention: Compared with the sham operation control group, relative volume ratio of bone trabecula, mean thickness of bony wall of bone trabecula and mean thickness of bony wall of cortieal substance of rats in the gonadectomy control group and low frequency pulse electric-magnetic field group obviously decreased (P 〈 0.05). Formation surface of bone trabecula, absorbing surface of bone trabecula, deposite rate of bone mineralization, percentage of cyclomycin labeled fluorescent perimeter, amount of osteoclast markedly increased (P 〈 0.05). ③Assaying findings of content of nitrogen oxide and activity of NOS in supernatant of serum and bone homogenate after magnetic field intervention: The content of nitrogen oxide and activity of NOS in low frequency pulse electric-magnetic field group remarkably increased as compared with the gonadectomy control group (P〈 0.05). CONCLUSION: Low frequency pulse electric-magnetic field can increase the content of nitrogen oxide and activity of NOS of ovarlectomized rats so as to inhibit bone resorption of osteoclast and reduce high conversion rate of bone remodeling.
