中文摘要:目的探讨重组热休克蛋白72(HSP72)对体外50%胎牛血清培养的大鼠肝细胞脂肪变性及肝细胞损伤的影响。方法 SD大鼠肝细胞分4组培养。①对照组:DMEM培养液(不含血清)与多粘菌素B;②实验组1:对照组培养液中加50%胎牛血清;③实验组2:对照组培养液中加50%胎牛血清与重组HSP72;④实验组3:对照组培养液中加重组HSP72。动态检测各组肝细胞内甘油三酯(TG)含量、脂肪变性率及悬液中ALT、AST含量。SPSS11.5软件统计分析实验结果。结果对照组肝细胞内TG含量及悬液中ALT、AST含量无显著变化(均P>0.05),肝细胞脂肪变性率为0。实验组1、2肝细胞脂肪变性明显,肝细胞内TG及悬液中ALT、AST含量较对照组均显著升高(均P<0.01),且与其它组比较实验组2变化更为显著(P<0.01);实验组3肝细胞内TG无明显变化(P>0.05),肝细胞脂肪变性率为0,但悬液中ALT、AST含量却较对照组有显著升高(均P<0.01)。结论重组HSP72能导致肝细胞损伤,但不能诱导肝细胞脂肪变性;重组HSP72能加重50%胎牛血清所诱导的肝细胞脂肪变性及肝细胞损伤。

外文摘要:Objective To explore the role of recombinant heat shock protein 72 (HSP72) in the steatosis and injury of rat hepatocytes in vitro with 50% fetal bovine serum culture. Methods SD rat hepatocytes were cultured in four culture so- lutions. Control group was with DMEM culture fluid( without serum) and Colistin B;Experimental group 1 was with 50% fetal bovine serum based on the solution of the control group;Experimental group 2 was with 50% fetal bovine serum and HSP72 based on the solution of the control group ; Experimental group 3 was with HSP72 based on the solution of the con- trol group. The levels of triglyceride(TG) in hepatocytes, the steatosis rates of culture hepatocytes, and the levels of ALT and AST in the culture suspension were detected timely. The experiment data was compared and analyzed by SPSS 11.5 software. Results The steatosis rates and levels of TG in hepatocytes, and the levels of ALT and AST in the culture sus- pension did not change significantly in the control group ( P 〉 0.05 ). After being cultured, the steatosis rates and the levels of TG in culture hepatoeytes, and the levels of ALT and AST in the cell culture suspension were markedly elevated in ex- perimental group 1 and experimental group 2 ( P 〈 0.01 vs control group), and that of more in experiment group 2 ( P 〈 0.01 vs. other groups). In experiment group 3 ,the hepatocytes steatosis rates and TG levels did not change significantly ( P 〉 0.05 ), but the levels of ALT and AST in the culture suspension were significantly higher than that of the control group( P 〈 0.01 ). Conclusion Recombinant HSP72 not only caused hepatocytes injury in vitro, but also increased 50% fetal bovine serum-induced hepatocytes steatosis and liver cell damage. However,recombinant HSP72 did not alone induce cultured hepatocytes steatosis.
