中文摘要:目的本研究基于网络药理学的研究策略,构建大黄蛭鳖汤“活性成分-靶点-通路”之间的关系,以探究该汤剂治疗骨髓纤维化的作用机制。方法通过TCMSP和TCMID数据库筛选出大黄蛭鳖汤中八味中药的主要活性成分,将所得成分通过PubChem平台和ChemMapper服务器预测其作用靶点。将预测的靶点和疾病相关靶点进行数据库归一化,并在Excel表格里进行映射,获得大黄蛭鳖汤的活性成分-作用靶点表,最后通过Cytoscape软件绘制蛋白作用靶点的化合物-靶点-疾病三者交互作用网络。结果大黄蛭鳖汤中共筛选出128个活性成分,涉及15个蛋白作用靶点。网络分析结果表明靶点主要参与细胞、组织、器官、代谢、免疫和应激等过程,通过调节TP53、FGF2、AKT1等信号通路来发挥治疗骨髓纤维化的作用。结论基于网络药理学的研究显示,大黄蛭鳖汤治疗骨髓纤维化具有多成分、多靶点的特点,为进一步研究大黄蛭鳖汤治疗骨髓纤维化的作用机制提供了新思路和新方法。
外文摘要:Objective Based on the research strategy of network pharmacology,this study constructed the relationship between"active ingredient-target-pathway"of Dahuang Zhibie decoction,in order to explore the mechanism of this decoction in treating myelofibrosis.Methods Through TCMSP and TCMID database,the main active components of eight kinds of traditional Chinese medicine in Dahuang Zhibie decoction were screened,and the obtained components were used to predict their targets through the PubChem platform and ChemMapper server.Normalize the predicted targets and disease-related targets in the database,and map them in the Excel table to obtain the active ingredient-action target table of the Dahuang Zhibie decoction,and finally the Cytoscape software was used to draw the compound of the protein action target-target-disease interaction network.Results A total of 128 active components were screened from Dahuang Zhibie decoction,involving 15 protein targets.The results of network analysis showed that the target was mainly involved in the process of cell,tissue,organ,metabolism,immunity and stress,and plays a role in the treatment of myelofibrosis by regulating TP53,FGF2,AKT1 and other signal pathways.Conclusion Research Based on network pharmacology shows that Dahuang Zhibie decoction has the characteristics of multi-component and multi-target in the treatment of bone marrow fibrosis,which provides new ideas and new methods for further research on the mechanism of Dahuang Zhibie decoction in treating bone marrow fibrosis.
