中文摘要:目的制备壳聚糖/盐酸小檗碱缓释微球。方法以壳聚糖为载体,通过SPG膜乳化技术制备缓释微球。以交联剂用量、混合搅拌时间、体系温度为影响因素,包封率为评价指标,星点设计-效应面法优化制备工艺。傅立叶变换红外光谱(FT-IR)、X射线衍射(XRD)、扫描电子显微镜(SEM)进行表征。膜透析法考察体外释药行为后,拟合动力学模型。结果最佳条件为交联剂用量8 mL,混合搅拌时间112 min,体系温度44℃,包封率80.6%,载药量13.89%。所得微球在2种pH值(1.2、7.4)下具有良好的缓释性能,释放均符合Peppas模型和non-Fickian扩散。结论该方法稳定可靠,可用于制备包封率高、缓释性能良好的壳聚糖/盐酸小檗碱缓释微球。

外文摘要:AIM To prepare chitosan/berberberine hydrochloride sustained-release microspheres.METHODS With chitosan as a carrier,the microspheres were prepared by SPG membrane emulsification technology.With cross-linking agent consumption,mixing time and system temperature as influencing factors,encapsulation efficiency as an evaluation index,the preparation process was optimized by central composite design-response surface method.The characterization was performed by Fourier transform-infrared spectroscopy(FT-IR),X-ray diffraction(XRD)and scanning electron microscopy(SEM).The in vitro drug release behaviors were investigated by membrane dialysis method,after which dynamic model was fitted.RESULTS The optimal conditions were determined to be 8 mL for cross-linking agent consumption,112 min for mixing time,and 44℃for system temperature,the encapsulation efficiency and drug loading were 80.6%and 13.89%,respectively.The obtained microspheres demonstrated good sustained-release performance at two pH values(1.2,7.4),whose release accorded with Peppas model and non-Fickian diffusion.CONCLUSION This stable and reliable method can be used for the preparation of chitosan/berberberine hydrochloride sustained-release microspheres with high encapsulation efficiency and good sustained-release performance.
