中文摘要：目的观察D2受体部分激动剂特麦角脲对线索诱发大鼠海洛因觅药行为复发的影响。方法建立大鼠海洛因固定比率自身给药行为模型,大鼠随机分为4组各8只:特麦角脲0.1mg/kg、0.2mg/kg、0.4mg/kg干预组(0.1mg/kg组、0.2mg/kg组、0.4mg/kg组)、生理盐水干预组(对照组)。戒断后第15d给予生理盐水或特麦角脲相应剂量腹腔注射,观察条件线索引燃海洛因觅药行为。结果①0.1mg/kg组各项行为指标与对照组相比,差异均无统计学意义(P>0.05);②0.2mg/kg组和0.4mg/kg组的总有效鼻触、第2h有效鼻触、获得条件刺激数均较对照组低(P<0.01);③大鼠活动度、无效鼻触各组之间差异均无统计学意义(P>0.05)。结论特麦角脲剂量依赖性地降低海洛因觅药行为重建,对运动功能无明显影响,有用于戒毒治疗的前景。

外文摘要：Objective To observe effects of terguride, a D2 receptor partial agonist, on cue-induced reinstatement of heroin-seeking behavior in rats. Methods Rat model of heroin self administration was first established with fixed ratio schedule, and were then randomly divided into 4 groups （ n = 8 ） ： saline treatment group and terguride 0. 1 mg/kg, 0. 2 mg/kg, 0. 4 mg/kg treatment groups. Rats received saline or different doses of tergnride i.p. treatment on 15th withdrawal day and cue induced heroin-seeking behavior was recorded. Results Compared with saline treatment, terguride 0. 1 mg/kg did not significantly modify the behavior response of cue induced relapse （ P 〉 0.05 ）. In contrast, terguride 0. 2 mg/kg or 0. 4 mg/kg significantly decreased the number of total nose poke, the 2nd hour nose poke and conditioned stimulus （ P 〈 0.01 ）. The difference in spontaneous activity and responses on control nose poke were not significant among four groups. Conclusions Terguride dose-dependently decreases cue induced heroin-seeking behavior, and does not affect spontaneous activity, suggesting that terguride may have potential therapeutic perspective for the treatment of drug addiction.
