中文摘要:目的:建立一种阿达帕林脂质体中药物含量的测定方法。方法:采用一阶导数紫外分光光度法,测定波长279 nm处的一阶导数峰-零谷值与阿达帕林脂质体中药物含量的线性关系。结果:在279 nm处一阶导数峰-零谷值可有效消除辅料磷脂对阿达帕林测定的干扰,阿达帕林在6.12~36.72μg.mL-1范围内,浓度与零谷值之间存在良好的线性关系,r=0.999 7;平均回收率为99.42%,RSD为0.72%(n=9);日内及日间精密度的RSD均小于1%(n=5)。经配对t检验,该方法的测定结果与用HPLC法测定结果没有显著性差异(P〉0.05)。结论:该方法简便、快速、结果准确,适于测定阿达帕林脂质体中药物含量。

外文摘要:Objective:To establish a method for determining the content of adapalene in liposomes.Methods:The first derivative UV spectrophotometry was used to determine the content of adapalene in liposome with wavelength at 279 nm.Results:The excipients in the liposome did not interfere with the assay.Linear relationship between the values of first derivative and the concentration of adapalene was obtained in the range of 6.12~36.72μg·mL-1(r=0.9997).The average recovery of samples was 99.42% with RSD 0.72%(n=9);RSD of the intra-and inter-day precision were lower than 1%(n=5).The results of first derivative spectrophotometry were not significantly different from those of HPLC method as analyzed by Paired-Samples T test.Conclusion:The method is simple,quick and accurate.It is suitable for determination of adapalene content in the liposomes.
