中文摘要:目的:探讨苏皖地区手足口病(HFMD)患者肠道病毒71型(EV71)和柯萨奇病毒A16型(CA16)分离株的VP1基因特征。方法:采集南京市、马鞍山市2009至2010年HFMD患者临床标本进行病毒分离和RT-PCR鉴定;扩增并测定VP1基因序列;从GenBank中选取EV71和CA16不同基因型参考毒株,利用生物信息学软件进行同源性比对和系统进化树分析。结果:共分离到9株EV71和4株CA16,EV71分离株的核苷酸和氨基酸序列同源性分别为96.5%~99.8%和99%~100%;CA16分离株的核苷酸和氨基酸序列同源性为96.2%~98.5%和99.3%~100%;系统进化树分析显示,9株EV71全部属于C4a基因亚型,而4株CA16则全部属于B1b基因亚型。结论:2009至2010年苏皖地区分离到的EV71和CA16毒株分别属于C4a和B1b基因亚型,与国内其他地区HFMD病原学及分子进化研究结果一致,有望成为疫苗候选株作进一步研究。
外文摘要:Objective: To investigate genetic characterization of the capsid protein VP1 of enterovirus 71(EV71) and coxsackievirus A16(CA16) strains.Methods: EV71 and CA16 strains were isolated using Vero cells from the anal and throat swab specimens collected from HFMD patients. The full-length VP1 genes of all isolated EV71 and CA16 strains were amplified by RT-PCR and then sequenced.The nucleotide and deduced amino acid sequences of VP1 gene were analyzed by biological software. Phylogenetic trees were constructed.Results: Nine EV71 strains and four CA16 strains were isolated from HFMD patients.The homogeneity of the 9 EV71 strains shared 96.5%-99.8% in nucleotide acid identity and 99%-100% in amino acid identity,and belonged to EV71 C4a subgenotype. The homogeneity of the 4 CA16 strains shared 96.2%-98.5% in nucleotide acid identity and 99.3%-100% in amino acid identity,and belonged to CA16 B1b subgenotype.Conclusion: These EV71 and CA16 strains isolated from Jiangsu and Anhui provinces in 2009 and 2010 belong to EV71 C4a and CA16 B1b subgenotype respectively,which are closely related to those isolated from other provinces in China.
