中文摘要:目的建立大鼠血浆中白求内酯Ⅱ和Ⅲ的HPLC检测方法。方法血浆样品经液-液萃取后,以乙腈-水(60:40)为流动相,流速1.0 mL·min^(-1)采用SHIMADZUODS柱(150mm×4.6 mm;i.d.,5μm)色谱柱分离,同时测定大鼠口服白术内酯Ⅱ和Ⅲ后血浆中两种有效成分的浓度。结果标准曲线线性范围为0.05~10.0 mg·L^(-1),线性关系良好(r>0.99),定量限为0.05 mg·L^(-1),回收率90%以上,日内和日间变异系数均低于10%。结论该法操作简便、灵敏、准确,适合于临床前药物动力学研究。

外文摘要:OBJECTIVE To develop a HPI.C-UV method for the simultaneous determination of atractylenolide Ⅱ and m in rat plasma. METHODS After a simple liquid-liquid extraction, atractylenolide Ⅱ , Ⅲ and the internal standard were separated on SHIMADZU C18 column(4.6 mm × 150 mm, 5μm) and detected by UV spectrometry.The detection wavelength was set at 237 nm. The mobile phase consisted of acetonitrile-water (60:40) at a flow rate of 1.0 mL·min^-1. The concentrations of atractylenolide Ⅱ and Ⅲ in rat plasma were simultaneously determined after oral administration by the method above. RESULTS The linear calibration curves were obtained in the concentration range of 0.05- 10.0 ms·L^- 1 (r 〉 0.99). The lowest limit of quantification was 0.05 mg·L^-1 . The recoveries were more than 90.0 % in plasma, and the intra- and inter-day coefficients of variation were less than 10.0 % in all cases. CONCLUSION The RP-HPLC method was specific, simple, sensitive and accurate. And the method was satisfied for the requirement of preliminarily pharmacokinetics study in rats.
