中文摘要:目的基于网络药理学和生物信息学的方法探讨抗骨痹方中4味主要中药(白花蛇舌草、半枝莲、山慈菇、蚤休)的活性成分、作用靶点以及抗多发性骨髓瘤(MM)的作用机制。方法采用TCMSP、BATMAN数据库分别获得白花蛇舌草、半枝莲、山慈菇、蚤休的活性成分和靶点,通过DisGeNET数据库获得MM相关靶点蛋白;采用软件Cytoscape构建成分-靶点网络和蛋白-蛋白相互作用网络,并对靶点进行了拓扑属性分析,运用其插件ClusterONE进行聚类分析,ClueGO进行通路富集分析。结果白花蛇舌草的活性成分有7个,半枝莲29个,山慈菇3个,蚤休4个。β-谷甾醇、豆甾醇、槲皮素等主要活性成分可作用于AKT激酶、磷脂酰肌醇-4,5-二磷酸-3-激酶、过氧化物合酶2等关键蛋白靶点,与调控细胞凋亡、神经营养因子信号通路以及慢性粒细胞白血病发生、发展等有关。结论抗骨痹方的主要4味药可通过调控细胞凋亡、抗血小板聚集等过程发挥多靶点、多通路的抗MM的作用。

外文摘要:Objective To investigate the active constituents,target sites and antimyeloma mechanism of four main herbs(Hedyotis diffusa,Scutellaria barbata,Cremastra appendiculata,and Paris polyphylla)in Kang Gubi Fang based on network pharmacology and bioinformatics.Methods The active components and target sites of Hedyotis diffusa,Scutellaria barbata,Cremastra appendiculata,and Paris polyphylla were obtained from TCMSP and BATMAN databases,respectively.Multiple myeloma related target proteins were obtained from DisGeNET database.The component-target network and protein-protein interaction network(PPI)were constructed by Cytoscape and the topological attributes of the target sites were analyzed.The cluster analysis was performed by using its plug-in ClusterONE and the pathway enrichment analysis was carried out by ClueGO.Results There were 7 active constituents in Hedyotis diffusa,29 in Scutellaria barbata,3 in Cremastra appendiculata and 4 in Paris polyphylla.Among them,β-sitosterol,stigmasterol and quercetin might act on key protein targets such as AKT kinase,phosphatidylinositol-4,5-bisphosphate-3-kinase,peroxidase synthase 2,etc,which were involved in the regulation of apoptosis,neurotrophic factor signaling pathway and chronic myeloid leukemia.Conclusion The main four herbs of Kang Gubi Fang can play the key role of multi-target and multi-pathway in anti-multiple myeloma through cell apoptosis,anti-platelet aggregation and other processes.
