中文摘要:目的探究苦碟子注射液对青春前期大鼠单侧睾丸扭转复位后生殖功能的远期保护作用。方法将32只4周龄健康雄性SD大鼠随机区组化分组法分为4组:假手术组、生理盐水组、苦碟子单次给药组和苦碟子连续给药组,每组8只。建立单侧睾丸扭转复位动物模型(720°2h)。于术后8周处死大鼠,检测睾丸组织内总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)活性、丙二醛(MDA)含量及Bcl-2/Bax蛋白的表达状况,计算Bax/Bcl-2比值。结果与同侧生理盐水组比较,两给药组SOD、T-AOC、NOS活性、Bel-2蛋白阳性表达率均有所增加,MDA含量、Bax蛋白阳性表达率减少,其中连续给药组差异明显(P〈O.05)及两给药组Bax/Bcl-2比值均显著下降(P〈0.05)。结论苦碟子注射液可通过有效清除氧自由基、保护氧化酶活性、抑制脂质过氧化,并通过促进Bcl-2蛋白、抑制Bax蛋白表达及调控Bax/Bcl-2的比值发挥抗细胞凋亡作用,从而对青春前期大鼠单侧睾丸扭转有明显的远期保护作用,且连续给药明显优于单次给药。
外文摘要:Objective To investigate the long-term protective effects of Kudiezi injection on genital function of immature rats following unilateral torsing/detorsing injury. Methods Thirty-two four-week-old healthy male SD rats were randomly divided into four groups: sham operation group, saline injection group, once injection of Kudiezi group and continuous injection of Kudiezi group (n = 8 for each group). Animals were submitted to unilateral 720 testicular torsion, then detorsion were done in two hours. Eight weeks later, the animals were sacrificed, total antioxidant capacity (T-AOC), superoxide dismutase (SOD), nitric oxide synthase (NOS) activity, malondialdehyde (MDA) content and the expressions of Bcl-2 and Bax protein were detected. Results Compared with saline injection group of the same side, the activity of T-AOC, SOD, NOS and the level of Bcl-2 protein were significantly increased in rats of Kudiezi injection groups. MDA content, the expressions of Bax and Bax/Bcl-2 rate were significantly decreased in continuous injection group (P〈0. 05). Conclusions Kudiezi injection protects the testis of immature rats following unilateral torsing/detorsing by clearing oxygen free radicals, protecting oxygenous enzyme, restraining lipid oxygenation, and reducing germ cell apoptosis by promoting Bcl-2 protein expression, restraining Bax protein expression and regulating Bax/Bcl-2 rate. There are also some protective effects on its contralateral testicular tissues from ischemia-reperfusion injury. The protective effect of the continuous injection is better than that of the once injection.
