中文摘要:目的:内质网应激与多种疾病相关,越来越多证据表明,内质网应激及其信号通路参与糖尿病的发病。资料来源:应用计算机检索1997/2004NCBIPubmed(www.ncbi.nlm.nih.gov)和中国期刊网数据库(www.cnki.net),选择检索对象为动物及人,检索词分别为endoplasmicreticulumstress(内质网应激)anddiabetes(糖尿病)。资料选择:根据检索词从两大数据库中检索中文、英文相关文献。资料提炼:检索到相关外文文献35篇,排除其中3篇日文综述;内质网应激中文文献6篇,未查及内质网应激与糖尿病关系的中文文献。32篇探讨内质网应激与糖尿病关系的英文文献中12篇为综述,余为论著;6篇中文文献均为关于内质网应激的综述。资料综合:对选取的文献进行阅读、归纳和分析。①内质网应激在多种疾病中起重要作用。低氧、低血糖、毒物等能干扰内质网功能,引起内质网蛋白合成增加而致蛋白过度负荷或错误折叠。②一定程度的内质网应激如分子伴侣表达能保护细胞免于损伤;若应激过强,保护机制不能与损伤相抗衡,则激活半胱氨酸天冬氨酸蛋白酶及生长静止/DNA损伤诱导蛋白-153等而诱致细胞发生凋亡。③内质网的稳态与胰岛β细胞功能状态相关,内质网应激既是细胞抵抗应激的生理反应,又是应激造成细胞损伤的重要机制。结论:胰岛β细胞含有丰富的内质网,是胰岛素的合成部位。分泌胰岛素的胰岛β细胞对内质网应激尤为敏感,胰腺内质网激酶-磷酸化细胞外信号调节激酶及其下游的真核细胞启动因子-2能调控胰岛β细胞的形成及其功能。

外文摘要:OBJECTIVE: Endoplasmic reticulum stress is associated with any diseases. More and more evidences prove that endoplasmic reticulum stress and its signal passage participate in the onset of diabetes mellitus. DATA SOURCES: A computer search of NCBI Pubmed(www.ncbi.nlm.nih.gov) and China National Knowledge Infrastructure (www.cnki.net) using keywords " endoplasmic reticulum stress and diabetes" in Chinese and English were performed to identify the related article published between 1997 and 2004. Studies with animals and human as subjects were selected. STUDY SELECTION: Rdated articles and reviews in Chinese and English were retrieved from two databases. DATA EXTRACTION: Thirty-five foreign language references were searched, excluding 3 Japanese references. Six Chinese reviews related to endoplasmic reticulum stress were searched. No reference on endoplasmic reticulum stress and diabetes was found in Chinese. Thirty-two English references related to endoplasmic reticulum stress and diabetes included 12 reviews and 20 articles. DATA SYNTHESIS: The selected articles and reviews were read, concluded and analyzed. ① Endoplasmic reticulum stress plays an important role in the onset of many diseases. Hypoxia, hypoglycemia and poison can disturb the function of endoplasmic reticulum, and lead to'the increase in protein synthesis so as to cause the overloading or wrong folding of protein. ② Endoplasmic reticulum stress to a certain extent can protect cells from injury, such as expression of molecular chaperones. However, if the stress is too strong, the protective mechanism cannot resist injury, and cell apoptosis can be caused by the injury of induced protein 153 through activation of aspartyl proteinase and DNA at static state. ③ There are 12 reviews and 20 articles in English references. Only 6 reviews were found in Chinese references. CONCLUSION: ER stress and the pathway of signal transduction play a prominent role in the development of diabetes.
